Temporal Profile of Reactive Astrocytes after Ischemic Stroke in Rats

چکیده
Ischemic stroke is a debilitating neurological disease most commonly resulting from an occlusion within the cerebral vasculature. Ischemia/reperfusion injury oftentimes consequence of stroke, characterized by oxidative stress, neuroinflammation, and activation surrounding glial cells following restoration blood supply. Astrocytes are regarded as prominent cell in brain and, under pathologic conditions, display, among other pathologies, activated (GFAP) relatively proportional to degree reactivity. The primary objective study was determine temporal profile astrocyte reactivity ischemic stroke. Thirty-four Sprague-Dawley rats were assigned surgery consisting either 90-min middle artery (MCAo) or sham surgery. Animals sub-grouped postoperative euthanization day; 2 days (n = 10), 4 11), 7 13). Fluorescence microscopy densitometry utilized quantify GFAP immunoreactivity, which indicated non-linear relationship ischemia/reperfusion. Results demonstrated substantially higher levels MCAo groups than sham, with peak being shown brains euthanized on day 4. These findings applicable future research, especially investigation interventions that target reactive astrocytes injury.

Notch signaling regulates nucleocytoplasmic Olig2 translocation in reactive astrocytes differentiation after ischemic stroke
Treatment with DAPT, an inhibitor of the Notch-activating enzyme, γ-secretase is known to reduce damage to ischemic brain. However, the molecular mechanisms supporting this therapeutic effect are not fully understood. Here we demonstrated that Notch/RBP-J signaling is activated in NG2(+) glial progenitors and reactive astrocytes such as GFAP(+) cells, Nestin(+) cells and RC2(+) cells, using Not...
متن کاملP1: Dextran Curcumin Promotes Novel Object Recognition Memory in Rats after Ischemic Stroke
Ischemic stroke causes the depletion of energy and induces excitotoxicity and neuroinflammation in the brain that results from thrombotic blockage. Cerebral ischemia leads to many types of memory loss, including impairment of working, spatial and object recognition memoreis. Curcumin shows strong anti-oxidoinflammatory activities but it terapathics limited by its low solubility in water and cor...
متن کاملEffects of Aerobic Training on Caspase-3 Expression and Apoptosis in Hippocampus of Rats after Brain Ischemic Stroke
Introduction: Although exercise is an effective strategy for preventing and treating stroke, the extent of this effect seems to depend on when exercise begins. Apoptosis plays a critical role after stroke. However, it is unclear whether early exercise inhibits apoptosis after stroke? The aim of this study was to determine the effect of eight weeks of early aerobic training after stroke inductio...
متن کاملTemporal profile of body temperature in acute ischemic stroke: relation to stroke severity and outcome
BACKGROUND Pyrexia after stroke (temperature ≥37.5°C) is associated with poor prognosis, but information on timing of body temperature changes and relationship to stroke severity and subtypes varies. METHODS We recruited patients with acute ischemic stroke, measured stroke severity, stroke subtype and recorded four-hourly tympanic (body) temperature readings from admission to 120 hours after ...
